One thing I think I remember seeing somewhere on here was a guy talking about his 4-of cube. You have a smaller cube and draft singleton style, but then during construction, you can use any of the cards you drafted as a 4-of. I don't remember if they did the usual 3 packs of 15 cards or if the decks were still 40 cards.
It'd be really tough to balance and choose what can't be used since you don't really want suked up vintage decks. Or maybe that is what you want.
Anyways, it seemed like an interesting format worth mentioning. Stand-outs I could see that are normally underwhelming in cube are Delver, Urza Lands and/or Cloudpost maybe, Aether Vial, Cabal Therapy, Blinkmoth Nexus.

I feel like the first thing, if any, that are added as multiples should be mana-fixing lands at higher Cube sizes. After that, cards to support certain archetypes that, for lack of a better term, suck unless you spike certain cards (like rituals/storm cards in storm or Urborg, Tomb of Yawgmoth/Corrupt-type cards in monoblack, maybe Urza lands?)
I don't think something as unique and powerful as Stoneforge Mystic should be a card to be included in multiples.
I call my library cube... so yes, cube should include multiple copies of a card.
Besides, if you want it for drafting; it is possible to open up multiple copies of a single card in a draft using sealed packs.
How many people do you think adhere to the singleton rule not because that's how they want to play, but rather that's how you are "supposed" to play?
Nobody. Everybody's cube is exactly the format they want it to be, and nothing more. People don't run cards or impose design restrictions because they're supposed to, they do it because that's how they want to design and play their cube.
How many versions of incinerate are needed for a 540 cube? Currently I run Incinerate, Lightning Strike and Searing Spear, but I am trying to make some hard cuts to get more good red spells in and am considering cutting one of these guys (probably Searing Spear). Thoughts on that situation?
I would probably try and play all 3 still, since 2cc splashable 3-damage instant-speed burn spells without targeting restrictions are pretty efficient. But ask your playgroup if they'd be willing to play less of them. If they won't miss the redundancy for that effect, it might be a better cut for your group than it would be for mine.
I was in the same spot as you recently and ended up cutting Lightning Strike to get Bonecrusher Giant in when Eldraine dropped. If anyone has an opinion on what else I am currently running that I should cut to get the strike back in, I would be very open to suggestions. (cube link in my signature).
