MaRo's wording is always suspect, but I don't think he'd mention Poison, which he knows has a particular connotation in Magic, if it wasn't coming back in some form. Especially since it will have been a full decade since it last showed up.
I do believe Maro was referring to poison counters. I think it will be something that interacts with poison counters, but doesn't actually make them. Like a proliferate or Deepglow Skate variant.
My point about the Cosmo Snake was that, just because it is venomous in lore, doesn't automatically mean poison counters. But it could.

Possibly. Depends on the color of the creature, I think. For example, a green creature might get "When this creature is blocked, the blocking creatures controller gains a poison counter" as a trample alternative. A black one might have a variant on the Sengir ability, where instead of getting bigger for being involved in creature death, it poisons the opponent.
Gonna call: the world serpent or a related card is able to directly put 10 poison counters on a player if a certain condition is met

How i feel about competitive players and casual players in EDH: The competitive are german tourists, the casual are italian tourists, both in a italian beach. The italians asking themselves "why are the germans here?" make a legitimate question, the answer is because the beach is beautiful, no matter the country you came from. The italians wanting to ban the germans are dumb, because if the germans pay for their stay and follow the rules like everyone else, they have the right to be in the beach. Hovewer, if the germans started to ask themselves "why are the italians here?"... they would be dumb as hell.

That's... not the right conclusion to draw? Pseudo-proliferate would still kill someone with poison counters. What his answer eliminates is the various guesses that were "target player loses" flavored as poison or poisoned weapons. It's going to be able to kill someone with poison counters, but that doesn't mean it will provide them itself.
Fight seems to be the most obvious one, although I could see a card-draw effect based off toughness rather than power as Green tends to usually use for effects like that.

Ok, so with Maro's clarifications, I would expect the poison card to either be:
* Something along the lines of Sabretooth Cobra for the Space Snake. It bites you once and the poison continues to build up.
* A BW Arrest variant (despictrd as being bound by serpents) that gives the victim's controller a poison counter every turn. Have to find a way to get rid of it quickly!
As far as a mistletoe reference, I expect it to be a way to remove indestructible creatures. Whether making them lose indestructible, exiling or -X/-X. Kills Baldr, but also those annoying Greek gods that are still hanging around.
I am wondering what color the snake will be. Dimir sultai or all 5 colors.
I wouldn't exclude the snake to do the 10 mana thing.
That is an interesting connection. We've already seen in Door to Nothingness that WWUUBBRRGGt is considered a reasonable cost for "Target player loses the game". So the could probably print "Target player gains 10 poison counters. (A player with 10 or more poison counters loses the game)" for the same cost. That would make it theoretically possible to kill someone with poison in limited, but mostly theoretical, and allow you to use a poison card with no other support (which was my biggest strike against actual poison counters, being that they need typically need to be a major theme of the set to do anything.
I honestly hope they don't do something like that... Just keep it simple. If the effect should kill somebody instantly than just write "Target player loses the game.". The art and name of the card will show you why he loses. Don't need to add additional layers and more text to a card. Poison was added as alternative win condition you need to build up. I would rather have an ETB effect of a really expensive creature that adds poison counters to players. Maybe 5 and you may choice how to distribute them or something like that. I don't know.
