My question came up the other day and internet searches have yielded very little as far as this exact issue is concerned.
Creature A is a 5/5 double strike, trample.
Creature B is a 4/4 Indestructible.
Creature A is on the attack and is blocked by creature B. My question here is the following:
Does creature B's toughness get reduced to 0 on the first strike damage and then creature A's full 5 regular strike damage go through? Or does creature B's toughness remains and creature A's regular strike damage is reduced by the 4 toughness again?
Unless the damage was dealt as infect or wither, the indestructable creature's toughness will not be reduced to 0. But what will happen here is during the first strike damage it can deal 4 damage to the indestructable creature, which is lethal damage to the creature and have the other 1 point of damage trample through. During the normal combat damage there is still lethal damage marked on the indestructable creature, so if you choose you can have it deal the full 5 damage to your opponent.
With trample you only have to assign lethal damage to a creature which is defined as damage equal to the creatures toughness.
With indestructible damage will still get marked on the creature until the EOT it just won't destroy the creature.
In your scenario your opponent ends up taking 6 damage 1 from the first strike and the full 5 in the regular combat damage step.
AS a final note normal damage never reduces toughness it is only marked and compared to it. To reduce a creatures toughness you need a source with wither or infect or a spell that specifically reduces it like infest.

When a creature is dealt damage its toughness is not reduced (unless the creature dealing damage has wither or infect, but then that's a consequence of a -1/-1 counter). Regular damage dealt to a creature is marked on that creature until end of turn, even for indestructible creatures.
The attacking creature must assign what would be lethal damage to its blockers during the first strike combat damage step; the blocking creature will then have at least 4 damage marked on it. During the regular combat damage step the attacking creature must again assign what would be lethal damage to its blockers. Difference is the one creature blocking it already has what would be lethal damage marked on it so the attacking creature can assign of its combat damage to the defending player.
Insofar as the attacking creature is concerned, the indestructible creature has 0 toughness (even though it doesn't). Good to know. This is something I myself have wondered (on and off) ever since Mirrodin.
