Kitchen table game with a friend, I have Leyline of Sanctity in play, he has Kalastria Highborn and a vampire dies from my blocker, can Kalastria Highborn's ability be activated for him to gain 2 life and I lose none? I told him no since it can't target me at all, but I rather have an official ruling, thanks.
Kalastria Highborn does not have any activated abilites just a triggered one. It will automatically go on the stack whenever a vampire goes into the graveyard, even if you have a Leyline of sanctity on the battlefield.
Also as the targetting condition for the Highborn is target player not target opponent he can always target himselft with it and not pay the B.

while the Leyline you have in play does not give you Shroud, it gives you essentially the same effect of Shroud.

702.16. Shroud
702.16a Shroud is a static ability. "Shroud" means "This permanent or player can't be the target of spells or abilities."
The word "target" is what makes Shroud and the Leyline beneficial to you.
Kalastria Highborne's ability targets a player, he must choose a target for the ability to hit.
Because Leyline says "you cannot be the target of spells or abilities an opponent controls" he cannot choose you as that target.
Sorry I should've been clearer, we know that he can't target me, and target himself is silly since it nulls the 2 life gained, but the confusion was that can he still gain 2 life and I lose none even if he can't target me, thanks.

While it may be "silly," it's actually required that he target himself. When a triggered ability with a target goes on the stack, a legal target must be chosen for the ability if one exists.
Of course, it's not required that he pay the mana to actually have that triggered ability do anything. But he must target himself if he can't target you.
