So last friday i was playing this treefolk deck. He had his banneret out, and was playing his Dauntlyss dourbark for 3 mana. In response i incinerated his Banneret. What happens to the land she tapped, to they mana burn him? Do they become untapped because he was unable to play the spell? Is his spell countered by this?
What happens? They all die in a fire. A very big fire. And do you know why? Because you didn't use card tags. Look at what you've done! Think of the poor little trees that will never know their parents because you murdered them by forgetting card tags yet again. -Woap

You do not get priority while your opponent is playing a spell, so you cannot burn out his banneret before he pays. If you want to prevent him from casting things using the discount from the banneret, you have to destroy it beforehand. Since most creatures can only be played at sorcery speed, you could kill the banneret during his upkeep or during your own turn.
"In response" to a spell would mean after that spell was on the stack and all costs had been paid. So it wouldn't change anything except prevent him from playing more things in the future.

Playing a spell involves several steps. One of the first of these steps is to determine the cost.
Printed cost + cost increasers - cost reducers = total cost.
The Bannaret reduces the cost to 2G, and once that's determined, it's "locked in" and nothing can change it.
