Once they select my creature enchanted with Pemmin's if I activate U: enchanted creature may not be the target of spells or abilities this turn'? Does it resolve or since it cannot be targeted does the sheltered by ghosts poof since it was cast then priority passed and during that time the target was declared ineligible by the activation'
Or do they get the card back to try again or to select another target as there was no eligible target for this?
When the opponent chooses your creature as the target of the Sheltered by Ghosts' ability, that is to say that they have put the triggered ability on the Stack and selected your creature as a target of that ability. You respond to this by activating "{U}: Enchanted creature gains shroud until end of turn." The ability gives the creature shroud. The aura spell sheltered by ghosts has only a single target, the creature your opponent wants to enchant. If you're fine with it, it resolves and enters the battlefield enchanting your opponent's creature. At this time, the second ability triggers and your opponent chooses a target to exile, in your case your creature.
You sure can respond to this ability by giving your creature shroud with pemmin's aura. If you do, your opponent's ability fizzles, it has no effect because the target is illegal and your creature isn't exiled. This does not remove the aura enchanting your opponent's creature which has still +1/+0, lifelink and ward. It has just failed to exile any permanent.
Note that you could have given shroud to your creature in response to the sheltered by ghosts spell instead. If you do that, your opponent hasn't chosen a target for the triggered ability yet. His aura will still enter the battlefield in this case and he won't be able to choose your creature as the target for the triggered ability. He will have to choose another permanent you control as the target, for example your pemmin's aura. This case is generally worse for you, so you may not want to do that. It's better to wait for your opponent to choose a target for the triggered ability of sheltered by ghosts, and if they do choose to target your creature, protect it in response to the ability.
