It was heavily implied by Rosewater that day/night would be coming in. His (not-verbatim) quote is: "Werewolves work exactly like you think". Considering that werewolves transform by the light of a moon, this statement of his, coupled with the "sun/moon" graphic on card pictures we've seen now, can easily be toted as a new mechanical function of some kind.

none because hardly any cards in general have been spoiled from the set yet. this is common knowledge, you aren't being a revolutionary by acting like you are the first person to think 'what if this day/night thing isn't a new mechanic' the thread is 80 pages, i'm sure it's been covered a good half a dozen times

...the only thing left now is how does it morph between Day and Night mechanic.
As far as we could read the blurry French, Howlpack Alpha said, "At the beginning of each upkeep, if a player cast 2 or more spells on the previous turn...(verb) Howlpack Alpha." Presumably, if nothing happens, it stays day/night, and if stuff happens, it becomes night/day. Interesting interaction with Ethersworn Canonist and Rule of Law, though...
So far, we've seen it on the Mayor/Alpha split, which was corroborated by the french card on the poster of Garruk, as well as the super-faded Cloistered Youth from the Youtube blur video. Sun/Moon on the first pair, Sun on the second. That's not really speaking of coincidence when it's from multiple different locales.

Nobody's saying the white card has to be a werewolf. It could be a girl who turns bansai super killer from being possessed by a geist that takes control at night, or it's a Jekyll/Hyde complex; who knows what sources they drew upon for the "horror" theme of the set?
The point is, Day/Night does not automatically mean Human/Werewolf, but they're probably going to be the primary benefactor of it, what with Innistrad having a minor tribal component to it. Tribal implies "mechanical identity", usually. Look at Lorwyn/Morningtide in how they handled that.

Alternative gameplay: Separate cards, drafted separately. But Day cards can be evolved into the Night cards (indicated by the dot next to the types rather than the symbol, maybe) whether you control the Day card or not. You control the card that you play, and you control it as long as that creature is in fact the one that's active. If you transform back, the other player controls its other form.
You can use the cards together, or you can use them interactively. Day cards will be very strong to account for the weakness, and the extremely narrow and unlikely (draft or constructed) situation of using both the Day and Night creatures yourself will be bolstered, because 'that linear trash' has always ended up undervalued in previous sets.
(oh, and, I think there may be three phases of time)

I think a lot of people are assuming it means that you can't play the card on its own. It has to be brought into play by the Mayor transforming.

maybe there will be cards that flip over, like they had in kamigawa. or maybe all day/night cards explain on them different ways to change the mode.
anyway, now we know that werewolves are green (and maybe red too)
We've known werewolves are Green and Red for a while. Other stuff you may have missed:
Vampires are Black and Red
Zombies are Blue and Black
Ghosts (creature type spirit) will show up in all colors, but more heavily in White and Blue.
Humans will also show up in all colors, but primarily Green and White.
These won't be Kamigawa style flip cards. The returning mechanic is Flashback. Day/Night is a separate mechanic that we haven't seen before, and don't yet know how it works. However, the best guess given what we've seen is that they will somehow be two-part cards. Two physical cards with the same collector's number ([number]a and [number]b) the night cards will have that darker boarder and most likely come in the same pack (since they have the same collecter's number) with the day version. Most likely they will somehow transform back and forth into eachother, which is one of the big things that separates them from kamigawa flip cards. (It's in the comprehensive rules that flip cards cannot flip back.)

My thought is something similar to the Ravens from A Game of Thrones TCG.
You have certain cards that expressly say 'It is Day/Night' to set the mode. They also get sacrificed if something else comes in that changes the day/night state. Its a very clear way to indicate the state.
Though god knows why we have two different cards for him.

The 163a is old news. The 234 is not. So that's where, like, 30 of the cards went... (Could have thought that there were "260+" cards in Innistrad.)
Me, it's either split cards or the Yu-Gi-Oh Extra Deck/Command Zone. (Oh, God, collectible card games are starting to become similar to each other. First, renaming "remove from the game" into "exile" (MTG) and "banish" (YGO), now this...)
It is 264 not 234. The previous poster saying 234 needs to check the picture of the mayor again.
My speculation:
1. The transformed versions are tokens... reference to the guys who saw that the set has 234 cards [the other 30 are 'b' version tokens]
2. The day night mechanic is triggered by opponent playing x number of spells per turn. Possible that there are three versions to reference harvest moon (play three spells to trigger/end), harvest moon (play two spells to trigger/end) and new moon (no trigger).
3. There should be a season card for players to reference - vampire nocturnus's mechanic is impossible if both players have conflicting cards turned over on top of their deck.
4. Flashback returns, partly because players can use flashback to assist in turning on / off the seasons.
5. Transformation occurs by exiling / sacrificing the "day" version and calling in the "night" version from exile / returning, and when season changes, vice versa.
6. The <3/3> on mayor may actually allow the mayor to turn into any 3/3 werewolf, though wizards will probably have only a few at each slot of 3/3, 4/4, 5/5, etc. Howlpack Alpha might be a "rare" token, but there are other common versions which may allow easier play for spirits, werewolves, and vampires that sleep in the day.
7. Cloistered Youth turns into a <blue> spirit at night. She's a vessel for spirits to possess and have phyiscal form. I was speculating a blue spirit "possesser" which etb by "championing" a creature on the battlefield in place of mind control... but there's evil twin already...
8. The silver colored card is Barren Glory or... AWOL.
9. Rusalkas return - I hope Verident Edelon comes back too.
Kamigawa-like flip cards would've made more sense than having two cards to go along with each other. does the extra card come in the pack? if so, it causes difficulty in drafting, because you'll have to take two cards. is that guy taking a day/night card, or is he just taking two cards? or maybe the card is printed on both sides, which means it would have no card back, which causes obvious problems, and would make sleeved games different from unsleeved games. the most logical thing would be to make it a token, and put it in the same pack as the card it goes with. But then they can only put one day/night card into each pack, because there's only room for one token.
