Obviously, Jimi Hendrix is at the top of the list. There just will never be another musician like him.
I'd love to see Pink Floyd, but I've seen Roger Waters. I don't know how much different it could be unless I hand-picked the set list (One of These Days, Echoes, Welcome to the Machine, all of Dark Side of the Moon, Sorrow...). That said, the Roger Waters show was one of the best concerts I've ever seen.
I'd really love to see Louis Armstrong, Miles Davis, Charlie Parker, and Billie Holiday, preferably in some NYC bar drinking with Jack Kerouac and Allen Ginsburg.
Aside from bands I'll never get to see, there are also bands I'd love to see at times that I can't see them. For example, I've seen Tori Amos many times, but to see her in 1992 when she was playing venues so small you could practically touch her piano must've been the greatest experience in the world. And I wish I had seen Fleetwood Mac on the Mirage Tour when they were performing a riveting version of Sisters of the Moon every night--it's one of my favorit Mac songs and I don't think they've played it since.
Rise Against, before Tim started going bald and got the crappy scene-hawk as a result, back when they weren't famous and they didn't touch an acoustic at all during a set, and they were gods among men. I refuse to go see them now because it'd reveal them as the pitiful lowly mortals they've become.
Pantera would've been cool, as would Metallica before Cliff died. Also not a band, but Tupac, hells yeah...

If there was just one band I could see live, it would totally have to be Mazzy Star. Not many remember them, but they've totally been my favourite band forever. Hope's voice is like that of an angel.

...that doesn't exist in their 'definitive' form anymore, (be it due to a death of a key member, break-up with no chance of reconciliation, whatever...) who would it be?
