It is triggered ability? When the 3rd +1/+1 counter resolved, could anybody response by destroying her so that she doesn't blow up?

Triggered abilities begin with "When," "Whenever," or "At," so this is not a triggered ability. It's simply part of the activated ability that gets performed only if the condition "this is the third time this ability has resolved this turn" is true at that time.
You can respond to the third activation by destroying Ashling. The ability will still resolve, but it won't do much of anything because there won't be an Ashling to remove counters from, so no damage will be dealt.

If someone responded to Ashling's third activation you could respond by activating it a fourth time which would resolve as the third activation of the turn, and thus remove all counters. Does this make sense?
