If you give the creature protection from red, yes. Combust can't be countered by spells or abilities, but it can be countered by game rules (for having an illegal target) just fine.
Yes you would if you declare the creature has protection from red. Whilst Combust damage can't be prevented part of protection is you can't be the target of any red spells so it would get countered on resolution.
