I am confused how the cleric's abilities actually work, like when they have prevent x amount of damage that would be dealt to target creature or player this turn. Like this card Kitsune Bonesetter
What, specifically, are you confused about? When Bonesetter's ability resolves, it creates a prevention effect. When a source attempts to deal damage to the creature or player, that effect prevents up to three of that damage from being dealt. The effect can prevent damage from multiple sources, up to a total maximum of three.

'This turn' and 'Until end of turn' effects wear off during the cleanup step, which is the last step of each turn. Thus, the effect will always end on the turn the ability was activated, whether it is yours or an opponent's.
And yes, you can activate such abilities during any player's turn, and at almost any time (specifically, whenever you have priority). Activated abilities can always be activated whenever you have priority unless they specifically state otherwise (e.g. Nezumi Bone-Reader).
I'm not sure exactly which point you want a ruling on, so here are all the relevant ones.
112.3b Activated abilities have a cost and an effect. They are written as "[Cost]: [Effect.] [Activation restriction (if any).]" A player may activate such an ability whenever he or she has priority. Doing so puts it on the stack, where it remains until it's countered, it resolves, or it otherwise leaves the stack. See rule 602, "Activating Activated Abilities."
