So I know there's been a lot of talk about the story of Innistrad, and what will happen this time around. Emrakul, Marit Lage, and even moon explosions have been mentioned. Although these are interesting, I want to stick to the facts. So, here's my theory.
We know the Angels go mad. We don't know how, but I can give a good speculation as to why. Angels were created by Sorin in order to help defend humanity on Innistrad, keeping the balance between monster and man. I believe that the Angels are driven mad by the fact that there is so much death and destruction on the plane, and their work is never finished.
The Angels come to a realization (somehow, maybe an outside influence) that there's only one way to end the madness: destroy the humans. Humans are the sole reason why you have Zombies, Werewolves, Spirits, and Vampires. Without humans, it leaves only the monsters to fight, and once they're all eliminated, there will be peace on Innistrad.
A comparison would be similar to how many science fiction novels relate technology and A.I. Coming to life in order to serve humanity, but then becoming self aware to the point where it realizes that humanity isn't good enough and must eradicate it for self-survival and a better world.
There are holes to this plot, yes: where do the demons fit into this? What about some of the other horrors of Innistrad? I'm sure the Angels want them dead, too.
Anyhow, it's just a theory I have that seemed to make sense.
The biggest obstacle to this is that it rules out any returns to Innistrad. Even if all the humans die but the monsters are not dead yet, WOTC would believe that a plane without humans would not be relateable. Because WOTC wants to keep popular planes around for future sets, this sort of massive, permanent change would be unlikely.
Only plane with zero humans that comes to mind is Lorwyn/Shadowmoor, and even if people like it, MaRo's mentioned more than a few times that a return there isn't too likely due, in part, to this.

What I mean to say is that based on that transform trigger, it would suggest that Avacyn feels probably regret or guilt or like a failure to protect someone/thing/whatever that she flips out and kills everyone instead. A bit... silly. I don't know how much I'll like this story direction...
Its extremely unlikely, but I'd like it if the uptick in demonic activity is what's pushed the Avacynian church in a more fanatical direction. The result of his being Avacyn taking a harsher hand against those elements. In doing so, you'd expect the fanatics to start winning converts to their side with stories of Avacyn's rebellion. Thus, facilitating Avacyn's shift into red mode, to do her damndest to cleanse her church of the fanatics and their poisoned sermons.
What's likely? Something silly like Eldrazi, or angry Nahiri.

Emrakul is slowly corrupting Innistrad's vampires - maybe she leaves and the corruption spreads onwards.
Nahiri is trying to prevent the corruption from spreading and killing the vampires.
Avacyn and the Angels are there to preserve balance: since we have less vampires (monsters), humans must die to balance it out.
The madness theme is the fact that everything was balanced and it's out of whack from the wrong side: monsters were never supposed to lose.
Delirium is the angels going haywire.
Sorin goes to innistrad to investigate and to his dismay, his angel is doing the wrong thing. He banishes her?
Jace finds clues leading to Tamiyo, together they try to find Sorin and they do.
