Edit: to clarify, with the Scepter you ARE still casting the spell, you just are not paying the casting cost. When you cast it, you have the choice of paying the kicker regardless of what you paid to cast the spell.
If you take a closer look at the rules text of Isochron Scepter, you'll see the copy of the spell is being cast. Because of that, you're allowed to apply optional additional costs like kicker (this is why Scepter-Chant is a thing).
Since it hasn't explicitly been mentioned, I'll say that you're NOT allowed to pay additional costs if you AREN'T casting the spell, let's say for example copying it with Fork. Isochron Sceptre works because you are actually casting the spell.

In saying that, I can still use Mana Leak too?
Not 100% clear what your question is, but you can do both of these things:
a) Imprint Mana Leak on the scepter, and cast it using the Scepter's ability; or
b) Cast Mana Leak targeting a spell that was cast using the Scepter's ability, because it was cast and it is a spell on the stack.
