No. Triggered abilities look for what actually happened, not for what tried to happen before replacement effects were applied. If Vizier causes an effect to not actually put any counters on a creature, then Flourishing Defenses won't see an event happen that triggers its ability.
If i were to put one -1/-1 counter on a creature i control, that was then reduced to 0 -1/-1 counters would this trigger Flourishing Defenses?
What you have here is two different abilities: One that is triggerd and one that replaces. So Flourishing defenses is constantly looking around for something to get a -1/-1 counter, and won't do anything until something does. Vizier, on the other hand, is more proactive. She sees that someone is about to receive a -1/-1 counter and intercepts that event and replaces it with another event. Only that replacement event actually happened though, so Flourishing Defenses didn't see any -1/-1 counters.
