Can you activate a vehicle's Crew ability once the vehicle itself has become a creature? And in situations like Veteran Motorist and such, are we allowed to use some other creatures to crew, say, Cultivator's Caravan, then on a later time on the same turn, tap the Veteran Motorist to grant +1/+1 on that Vehicle?
Or for a better example: Suppose you have Veteran Motorist, whose ability gives +1/+1 to the vehicle it crews (say Sky Skiff, and assume these two cards are the only nonland permanents I have). If I tap Motorist to crew a Skiff and give it +1/+1, then I untap Motorist somehow, can I tap the Motorist again to give the Skiff a second +1/+1 boost?
You can activate a vehicle's crew ability as many times as you are able to pay the cost though each additional time will usually have no real effect. In the cases you mentioned are the exception to usually having no effect, so yes you can do it.

If, hypothetically, there is a vehicle that has ability that says "Whenever this becomes crewed, it deals 1 damage to target creature or player", will that effect trigger repeatedly also?

Correct. It is similar to activating the equip ability of an equipment already on something. Even the nothing appears to change in the game anything that triggers from an ability being activated or something being targetted would still happen.
If I crew Cultivator's Caravan, and my opponent hits it with Grasp of Darkness, could I crew it again to reset it to 5/5? I feel like the answer is no, that the -4/-4 will still be applied, but I'm not certain.
No, the Caravan stays a 1/1. The Caravan gets its printed power/toughness when it is crewed, because only creatures can have p/t. But it doesn't actually set the p/t to any specific value. Just like with any other creature, the printed value is just the starting point from where you calculate the current p/t. Any modifications to it will apply over it.
It's worth noting that even if an effect did set the Caravan's power and toughness to 5/5 directly - for the sake of example, let's say that the Caravan is instead a Phyrexian Totem and its controller activates the animation ability a second time after it has been hit with Grasp of Darkness - it would still be a 1/1. Continuous effects are applied in a layer system, and effects that modify a creature's power and toughness will always apply after effects that set a creature's base power and toughness.
