Would it resolve in a sense that Vines would trigger first therefore not being able to be targeted by said spell? If not what is the sense of this card if every time I go to play it I get hit with an in-response kill the "un target able" creature. Besides the +4 +4
The stack always empty from the top and down. When you cast vines you are putting that on the stack, then an opponent responds with a bolt or something to kill the agent putting that on the stack on top of your vines.
Stack:
TOP
Bolt
Vines
BOTTOM
Those spells will then resolve form the top down. The bol resolves first, killing your blighted agent, after that your vines ill try to resolve, have no legal target and because of that get countered by the game rules.
If you want vines to resolve first you need to cast it after an opponent tries to remove your agent.
Basically, instead of your opponent casting a bolt in response to the vines you need to wait until an oppoent tries to kill your agent and then cast the vines in response to that. That would mean the vines resolves first, giving your agent "hexproof" makign the opponents removal fizzle when it tries to resolve due to the target being illegal.

The scenario I have ran into about 3 or 4 times now is the following.
My turn starts I target Blighted Agent with Vines Of Vastwood.
Opp. Lightning bolt Blighted Agent in response, or path to exile.. etc etc
