If I have a diffusion sliver on the battlefield, an I were to cast sliver construct, for example, and an opponent countered the construct, would diffusion sliver's ability take effect?

To answer your question, no if a spell or ability mentions a permanent type like creature it only refers to objects of that type whilst it is on the battlefield. Since your opponent is attempting to counter your sliver construct it has to be on the stack so there isn't anything for the diffusion sliver's ability to fire off.
