As in, your opponent saw you hand, and chose a card for you to discard? No. By then, it is too late as the card, lets say Duress for example, has already started resolving.
If you did it before you revealed your hand, you could activate the Scepter in response and pick a card to keep safe
W may only be paid with white mana. U may only be paid with blue mana. B may only be paid with black mana. R may only be paid with red mana. G may only be paid with green mana. C may only be paid with colorless mana. 1 may be paid with white, blue, black, red, green, or clolorless mana.
No sir. Once the ability resolves and the card is chosen, you can't do anything until after the ability resolves, because you can't interrupt an ability in the middle of it resolving. (That's rude!)
You can however, while the discard ability is on the stack, activate the ability of the scepter before the card is chosen. So, you can save your most valuable card, or whatever card you don't want to lose, but you will still have to discard a card.
Unless that card is the last card in your hand, you can't use the scepter to save you from having to discard.
You can't use it to stop you from discarding a specific card once it has been chosen.

Whats the big deal about black lotus you ask? Well you see, there is no big deal about it. It IS the big deal.
You can certainly use the Scepter in response to a discard spell targeting you. However, they would still get to look at your hand and choose a different card. Once you know what they are going to choose it would be too late to use the Scepter.
