Tough call... definitely one of the cards I've received as a gift from my wife and son. It would be between ROE Gideon Jura (cracked from a Father's Day pack) or DST Sword of Fire and Ice, given to me as a single to complete the cycle of swords.
Island. (I don't believe I am mistaken, but it could be one of the other three Islands from Invasion.)
The first card I possessed and a card that would be the basis of many of my decks. I don't think I would retain the exact card (which I marked) that I was given by a then friend of mine (drifted apart), but I would keep an Island with same illustration.
For others, the choice of card might depend on the reason for divesting yourself of all but one card. For me, however, the only card that I am particularly (or remotely) attached to is Island; the other cards involved much buying or trading, but that was either for the sake of play or collecting.
I sold my collection not too long ago and kept a few cards. Of them, if I had to choose one, it would be my FBB Italian Triskileon. I played CounterOath WAAAAAAY back in the day and I recurred this bad boy so many times, I just can't see myself parting with it

I'm on my third collection, so my first card is long gone (it was a Portal Denizen of the Deep that I'd give almost anything to have back). The card I'd keep today is my custom full art Necropotence. It was a gift to me in 2009, from someone I loved with all my heart, and though we've drifted apart and not spoken in many years, it reminds me of what we once had together. I'll never get rid of that card, not ever.
Unfortunately, over the years, I've lost or had stolen more than a few cards with sentimental value to me. But they keep getting replaced with continually more awesome ones. There's The Tabernacle at Pendrell Vale that I saved up for for months to finally purchase last year, even though I've wanted one off and on since 1995. There's the Avatar of Woe that RK Post altered for me while we got drunk at a local bar. And then there's the Betty Page Tempest Dark Ritual playset my wife commissioned Ken Meyer Jr. to do for me......it's a terrible choice I think I'd have to keep all of them, even if it meant not eating for a week or whatever disaster was befalling me to cause me to have to sell.

My Bazaar of Baghdad. Reserve list +300 bucks and its in near perfect condition. I would keep all my reserve list cards though. I think I own 4 that are worth any money.
Tough Choice. I can narrow it down to three, but one?
Foil Sword of Light and Shadow: Cracked this baby at my very first pre-release. It was Darksteel and my second tournament ever. The kicker? Mark Zug was there signing cards. That thing helped me get 4-1 and cement my love of this game. Sentimental doesn't even begin to cover it.
Judge Promo Balance: My then-girlfriend bought this for me at our first GP together. It was for my Birthday. We've have gone on to more GP's and she's my wife now. Yes, she plays. If only I could get her to stop rofl-stomping me with her Teferi EDH deck.....
Foil Akroma, Angel of Wrath: You know that one card you have that you used to smash face all the time, your friends were INCREDIBLY jealous of, and just made life that much better for you because you pulled it when you had a few bucks left before payday and were having a really, really bad day? Yeah, that's this one. She may not be worth as much as back in the day, but she's still shiny and she still feels good to lay on the battlefield and watch opponents 'oooo' at the pretty.
However, if I had to stick with only one......probably the Akroma. Not sure why. She just is.

I love my Drana EDH deck and the 3D is pretty much irreplaceable but I love U/B and wanted an Underground Sea for many, many years before I paid $100+ for one.
I paid a lot more for the dual land so I think I'd keep that. Barely.
Probably the Shadow of Doubt my friend and I signed after scrubbing out of 2HG states. It's the most tangible memory a card of mine possesses, since it's directly related to one tournament in my mind.
I'd say The Abyss, but I already gave it away as a Christmas gift to a friend, framed and all. I guess I will go with the Lord of the Pit, because awesome art is awesome.
My Sliver Queen is the one I'd keep. My favorite creature tribe, all the mana symbols for nostalgia, the original gold frame. Lotta good memories with that card.
Screw monetary value, it would either be: Crush of Wurms: First card I fell in love with and thought was overpowered. Monkey Cage: My favorite card of all time Graceful Adept (Foil): My favorite MTG art.
For what it's worth, the opposite of evergreen is "deciduous" so I suggest we start using that from now on to refer to shroud, banding, islandhome, etc.
Counterspell, if I had understood Magic as well as I do today, I probably would've owned a few Force of Will and that would have been my answer to selling all my cards with the exception to one(or at least a playset).
There's no way I could keep only one card. If I would be forced into selling, because financial situation, it would not be a beta Black Lotus or any other P9 piece, nor duals. Nor any of the more expensive promos, altough I would love it to be one judge Lightning Bolt, which was the first judge promo when I started judging. I would try my utmost to keep the Unhinged foil set, as it's getting very tedious to piece together nowadays.
Most likely it would be one 5k box of bulk commons, that sits at my office at work, which I use for protective cards, whenever I send out cards without toploader. Or maybe some common that was used as a bookmark and forgotten inside a book. (In that case it would be propably 10+ cards.)
I do have some signed cards I have a strong bond with, along with some artist sketches, that most likely have some monetary value, but I would prefer to keep, if I could. But I've been playing for over 19 years now, so there are lot of cards that have great memories. Choosing just one would be ridiculously hard.
But as the OP asked what would be the one card. Moneywise it would have to be the Lotus, altough it's my second beta Lotus. I had to sell the first one, just before graduating to buy a new fridge and cover some bills, since old bridge broke down and I missed money by one round at PT Kobe. So intellectually I know I can just buy a new one, if I needed it later (and the last time was a nice upgarde in condition. This is actually my fifth Lotus, three have been from Unlimited.)
My sentimental choice is either the judge Bolt or a Summer Grizzly Bears, that's in my global Grizzlies collection. Or maybe just a signed Camel. Card I've been collecting on and off for nearly 15 years.
It's drawn on a peeled foil. John was very nice about it and used to play some MtG back in the days. It is a 'tad' overpowered, but it's supposed to be a super mythic (in the Dork Tower comic circa 2006).
I have a strong bond with my foil Kataki's War Wage and foil Thoughtseize.
Probably would keep the Thoughtseize for it's impact on the game as a card.
I would like to say I would part with everything except one copy of each shock, a voice of resurgence, promo nightveil spectre, foil legion loyalist (favorite art!), my set of scavenging ooze and the first mutavault I pulled.
